Peptide stability is directly affected by temperature, light, oxygen, and moisture. Improper storage leads to degradation — oxidation of sensitive residues, hydrolysis of peptide bonds, and aggregation — all of which reduce biological activity and research validity. This guide covers the specific storage requirements for lyophilized and reconstituted peptides used in research.

Lyophilized Peptide Storage#

Lyophilized (freeze-dried) peptides are significantly more stable than solutions:

Storage temperatureExpected stability
-80°C (ultra-low freezer)5+ years; maximum stability
-20°C (standard freezer)2–3 years; standard practice
2–8°C (refrigerator)6–12 months depending on compound
15–25°C (room temperature)Weeks to months; not recommended for storage

Why lyophilized is more stable:

  • No water present → no hydrolysis (peptide bond cleavage)
  • No aqueous medium → no oxidative degradation pathways
  • Dessicated cake → no Maillard browning reactions
  • Reduced molecular mobility at low temperature → less structural degradation

Desiccant packs: Many suppliers include silica gel desiccant packs in packaging. Keep the desiccant in the storage container — it absorbs any residual moisture that enters when the container is opened.

Light Sensitivity#

Peptides sensitive to light (UV degradation):

Peptides containing the following residues are photosensitive:

  • Tryptophan (Trp, W): UV-sensitive; oxidizes to kynurenine under UV exposure
  • Tyrosine (Tyr, Y): Less sensitive but still photo-degradable
  • Phenylalanine (Phe, F): Minimal sensitivity
  • Cysteine/Met-containing: Oxidation of sulfur-containing residues under UV + oxygen

Common photosensitive research peptides:

  • Ipamorelin (contains D-2-Nal — naphthalene, which is UV-absorbing and photosensitive)
  • GHRP-2, GHRP-6 (aromatic residues)
  • HGH Fragment 176-191 (contains Trp)
  • PT-141 / Bremelanotide (His-D-Phe core)

Storage recommendation: Keep lyophilized peptides in amber vials (or wrapped in foil) away from direct light. Standard freezer storage already eliminates the most damaging UV exposure (freezers are opaque).

Reconstituted Peptide Storage#

Once dissolved in BAC water (bacteriostatic water):

PeptideReconstituted stability (2–8°C)Notes
BPC-15728 daysIn BAC water; amber vial recommended
TB-50028 daysSimilar to BPC-157
Ipamorelin21–28 daysSensitive to oxidation; use amber vial
GHRP-2, GHRP-621 daysAromatic residues; light-sensitive
CJC-1295 (no DAC)21 daysKeep cold; avoid light
Semaglutide28 days in sterile waterUse sterile saline or water (not BAC water)

Why BAC water extends stability: Benzyl alcohol (0.9%) is an antimicrobial preservative that prevents bacterial growth in reconstituted peptide solutions. Without it (using sterile water), microbial contamination can degrade the peptide within 24–72 hours.

Freeze-Thaw Cycles: Management#

What happens during freeze-thaw:

  • Ice crystals form during freezing → physical stress on peptide structure
  • Thawing → peptide may not fully re-dissolve (aggregation)
  • Each cycle reduces peptide integrity by some amount (compound-dependent)

Best practice:

  1. After reconstitution, divide into single-use aliquots (e.g., 200–250 μL volumes in small tubes)
  2. Freeze aliquots at -20°C
  3. Thaw only the volume needed for each experiment
  4. Never refreeze a thawed aliquot
  5. Label aliquots with date and concentration

Maximum recommended freeze-thaw cycles:

  • Most research peptides: ≤3 cycles
  • Sensitive peptides (disulfide-containing, aggregation-prone): ≤2 cycles

Shipping and Cold Chain#

What to do when peptides arrive:

  1. Check packaging integrity — vials should be intact with no cracks
  2. Check vial cap seals — no evidence of tampering
  3. Check lyophilized powder appearance — white/off-white cohesive cake (not loose powder or yellow)
  4. If compound arrived at ambient temperature (acceptable for lyophilized): inspect visual quality; lyophilized peptides tolerate 24–72h ambient transit without significant degradation
  5. Transfer to appropriate storage temperature immediately upon receipt
