Most injectable anabolic research compounds are formulated as oil-based solutions (typically sesame oil, MCT oil, or castor oil with benzyl alcohol/benzyl benzoate as co-solvents). However, several compounds exist as water-based formulations — either because esterification is impractical (stanozolol suspension) or to achieve rapid onset without an ester (testosterone suspension). Water-based formulations have distinct pharmacokinetic, stability, and injection-site characteristics compared to oil-based depots.
Oil-Based vs. Water-Based Injectables#
| Property | Oil-based (esterified) | Water-based (suspension/solution) |
|---|---|---|
| Release mechanism | Slow ester hydrolysis from oil depot | Rapid dissolution of crystals/direct absorption |
| t½ | Days to weeks (ester-dependent) | Hours to days (dissolution-limited) |
| Injection pain | Moderate (oil viscosity) | Higher (microcrystal irritation at site) |
| Injection site reaction | Mild oil depot; resolves in days | Often more local inflammation |
| Needle gauge | 22–23G typically | 22–25G (suspension may clog finer gauges) |
| Formulation stability | Months–years at RT | Shorter; crystalline can settle; shake before use |
Testosterone Suspension (No Ester)#
Composition: Testosterone micronized crystals suspended in sterile water (usually with polysorbate 80 or other surfactant)
Pharmacokinetics:
- No ester → no ester hydrolysis step → absorbed directly from injection site
- Onset: very rapid (~2–4h to peak)
- t½: ~24h (short; requires daily injection for stable levels)
- Advantage for research: near-immediate hormone exposure; no ester delay
- Disadvantage: daily injections; painful injection site reactions
Research application: Testosterone suspension is ideal when rapid onset is needed and daily injection schedule is acceptable — e.g., acute endocrine response studies, single-dose pharmacokinetic experiments.
Stanozolol Aqueous Suspension#
Composition: Micronized stanozolol crystals (particle size ~5–20 µm) in sterile water with benzyl alcohol (preservative) and surfactants
Why an aqueous suspension:
- Stanozolol cannot be efficiently esterified at C17 due to pyrazole ring steric effects
- Aqueous suspension provides the only injectable option for stanozolol
Pharmacokinetics:
- Dissolution-limited absorption: crystal particle size determines rate
- t½: 24–48h (broader range due to particle size variability)
- Peak: biphasic — initial burst from small crystals, prolonged tail from larger crystals
- Frequency: every other day (EOD) for relatively stable levels
Injection technique note: Aqueous suspensions settle on standing. Vial must be thoroughly shaken/swirled immediately before drawing dose. Failure to resuspend results in dose variability (early dose: water-heavy; later dose: crystal-heavy).
Winstrol Depot: Classic Aqueous Stanozolol#
The original Winstrol Depot (Winthrop/Sterling) was an aqueous stanozolol suspension at 50 mg/mL. Key characteristics still studied in research:
- Bioavailability: 100% (bypasses first-pass, no ester hydrolysis required)
- Hepatic exposure: significantly lower peak hepatic concentration vs. oral route → less BSEP inhibition per systemic unit
- SHBG suppression: less than oral (less first-pass hepatic AR activation)
- Anabolic effect: equivalent to oral at matched systemic AUC
Formulation Stability for Research#
Aqueous suspension stability:
- Settle on standing: always re-suspend immediately before use
- Temperature: do not freeze (ice crystals → particle aggregation → dose inconsistency)
- Shelf life: typically 18–24 months unopened; use within 28 days after first use (if preserved with benzyl alcohol)
- Light sensitive: store in dark/amber vials
Oil-based solution stability:
- More stable than aqueous: fatty acid ester protects compound from hydrolysis
- Can be stored at room temperature 2+ years if sealed
- After opening: stable for 6–12 months at room temperature; refrigeration extends
