Semaglutide is a glucagon-like peptide-1 (GLP-1) receptor agonist with 94% sequence homology to human GLP-1, modified at positions 8 and 34 to resist DPP-4 degradation and with a C18 fatty acid chain that binds albumin to extend half-life to approximately 7 days. The active compound in Ozempic (2 mg/week) and Wegovy (2.4 mg/week). Mechanism: stimulates glucose-dependent insulin secretion, suppresses glucagon, delays gastric emptying and acts on hypothalamic GLP-1 receptors to reduce appetite and caloric intake.
