MOTS-C (Mitochondrial Open Reading Frame of the 12S rRNA Type-C) is a 16-amino-acid peptide uniquely encoded by the mitochondrial genome — making it the first 'mitochondrial hormone' identified. It is produced in mitochondria and translocates to the nucleus to regulate nuclear gene expression, creating a retrograde mitochondria-to-nucleus communication pathway. Primary research findings: MOTS-C activates AMPK and FOXO transcription factors to improve insulin sensitivity (reducing diet-induced insulin resistance in rodents), enhances glucose uptake in skeletal muscle independent of insulin, increases endurance capacity, and extends lifespan in aging mouse models by 6 weeks.
